Desenvolvido para Xbox 360, PS3 e PC, Prototype remete à experiência obtida em Crackdown pois o protagonista passeia por prédios e construções lembrando as acrobacias corporais do esporte Parkour e os saltos daquele game. A trama se passa em Nova York onde o personagem principal está em busca de seu passado e dos mistérios que permeiam seus super-poderes, notadamente, o de absorver as memórias de quem ele matou e a capacidade de executar formas de ataques de maneira sobrehumana, além de poder se disfarçar como quem foi consumido por ele.
Dentre as categorias de mutação estão: disfarce, ataque, defesa e sentidos. O disfarce permite que o protagonista, chamado Alex Mercer, adquira a forma de alguém de seu interesse para usufruir de locais e privilégios que só ela tem acesso. Ademais, os poderes e conhecimentos desta pessoa também são obtidos por Alex, possibilitando centenas de combinações. Portanto, Alex Mercer está em busca de seu passado e sua história e irá, disfarçado sob as mais diferentes formas, perseguir sua memória através de sangue e explosão.
